چکیده

ARTICLE HISTORY Received 8 May 2017 Accepted 15 May 2017 ABSTRACT Much contemporary evidence underscores the pathophysiological importance of Ca2C handling by acidic organelles such as lysosomes. Whereas our knowledge of how Ca2C is released from these acidic Ca2C stores (the ‘outs’) is advancing, we know relatively little about how Ca2C uptake is effected (the ‘ins’). Here I highlight new work identifying animal Ca2C/HC (CAX) exchangers that localize to acidic organelles, mediate Ca2C uptake and regulate cell migration in vivo. Continued molecular definition of the acidic Ca2C store toolkit provides new insight into Ca2C-dependent function.

New aspects of nuclear calcium signalling.

Nuclear calcium signalling has been a controversial battlefield for many years and the question of how permeable the nuclear pore complexes (NPCs) are to Ca2+ has been the subject of a particularly hot dispute. Recent data from isolated nuclei suggest that the NPCs are open even after depletion of the Ca2+ store in the nuclear envelope. Other research has suggested that a new Ca2+ -releasing me...

Ca2+/H+ exchange by acidic organelles regulates cell migration in vivo

Increasing evidence implicates Ca(2+) in the control of cell migration. However, the underlying mechanisms are incompletely understood. Acidic Ca(2+) stores are fast emerging as signaling centers. But how Ca(2+) is taken up by these organelles in metazoans and the physiological relevance for migration is unclear. Here, we identify a vertebrate Ca(2+)/H(+)exchanger (CAX) as part of a widespread ...

Inositol 1,4,5-trisphosphate-induced calcium release in the organelle layers of the stratified, intact egg of Xenopus laevis

Using double-barreled, Ca2(+)-sensitive microelectrodes, we have examined the characteristics of the Ca2+ release by inositol 1,4,5-trisphosphate (Ins(1,4,5)P3) in the various layers of Xenopus laevis eggs in which the organelles had been stratified by centrifugation. Centrifugation of living eggs stratifies the organelles yet retains them in the normal cytoplasmic milieu. The local increase in...

cAMP-dependent recruitment of acidic organelles for Ca2+ signaling in the salivary gland.

Autonomic neural activation of intracellular Ca2+ release in parotid acinar cells induces the secretion of the fluid and protein components of primary saliva critical for maintaining overall oral homeostasis. In the current study, we profiled the role of acidic organelles in shaping the Ca2+ signals of parotid acini using a variety of imaging and pharmacological approaches. Results demonstrate ...

myo-Inositol 1,4,5-trisphosphate. A second messenger for the hormonal mobilization of intracellular Ca2+ in liver.

The stimulation of hepatocytes by alpha 1-adrenergic agonists and vasoactive peptides results in a mobilization of intracellular Ca2+ which is accompanied by breakdown of phosphatidylinositol 4,5-bisphosphate to release myo-inositol 1,4,5-trisphosphate (Ins(1,4,5)P3). The possible involvement of Ins(1,4,5)P3 in intracellular Ca2+ mobilization was tested using a preparation of saponin-permeabili...
